Use the NEB Tm Calculator to estimate an appropriate annealing temperature when using NEB PCR products.
Instructions
- Select the product group of the polymerase or kit you plan to use.
- Select the polymerase or kit from the list of products.
- If needed, modify the recommended primer concentration.
- Enter primer sequences (with up to 3 ambiguous bases). Spaces allowed.
Note that an annealing temperature will only be displayed if both primer sequences are entered.

- Enter primer sequence pairs (max 3 ambiguities/seq). Spaces allowed.
- Primer pairs (one pair per line) are expected to be in the format ID1 Primer1 [ID2] Primer2, separated by semicolons or commas. Data pasted or loaded from a file can also use tabs. ID2 is optional.
- Interleaved primer pairs (one primer per line) are expected to be in the format ID1 Primer1, separated by semicolons or commas. Data pasted or loaded from a file can also use tabs.
Results can be downloaded in tab-delimited format as a plain text file in many modern browsers. Input lines that do not match the expected format are ignored. Please visit Help for more information.
